As part of preclinical vaccine development, particularly for HIV, Indian rhesus macaques (Macaca mulatta) are used to evaluate antibody-based correlates of vaccine-mediated protection1. Four IgG subclasses are present in rhesus, but subclass numbering between rhesus and humans is not strictly consistent between species. Based on sequence alignment of the CH2 region responsible for FcγR recognition, rhesus subclasses are most similar to human IgG1. Most substitutions between rhesus and humans are located distal to the FcγR contact residues, except for H38 which borders an FcγR contact residue and is substituted to Q in all rhesus subclasses. Additionally, rhesus IgG2 has a deletion of P2, a residue proximal to the FcγR contact residues. Rhesus IgG2 binds to FcγR-conjugated fluorescently coded microspheres in a glycosylation dependent manner. Rhesus IgG2 and IgG3 can be specifically detected using antibodies raised against their hinges. In serum, rhesus IgG2 is the second most abundant Ig subclass, after IgG1. 3C10.1 was generated by immunizing mice with overlapping peptides derived from the IgG hinge region, VVHEPSNTKVDKTVGLPCRSTCPPCP and DKTVGLPCRSTCPPCPAELLGCPSVF1. Administration of antibody to rhesus macaques results in a moderate reduction of CD8+ T cells in blood.
