Description

Endotoxin, also known as lipopolysaccharide (LPS), is a crucial component of the outer membrane of Gram-negative bacteria. This complex molecule is found in common bacterial species such as Escherichia coli, Salmonella, Shigella, Pseudomonas, Neisseria, Haemophilus influenzae, Bordetella pertussis, and Vibrio cholerae. Understanding endotoxin is paramount in various scientific and medical fields due to its significant biological activity. The structure of LPS is tripartite: - Lipid A: This lipid component is primarily responsible for the toxicity and biological effects of endotoxin. It triggers potent inflammatory responses in mammalian systems. - Core Oligosaccharide: A short chain of sugars connecting Lipid A to the O-antigen. - O-Antigen (O polysaccharide): These highly variable polysaccharide chains are the cell wall antigens of Gram-negative bacteria. They are responsible for the immunogenicity of LPS and are often used for serological typing of bacteria. Furthermore, endotoxin (LPS) is a powerful stimulant of the innate immune system. It elicits a wide range of inflammatory responses, including the release of cytokines, chemokines, and other mediators, which can lead to fever, shock, and organ damage at high concentrations. Furthermore, LPS efficiently activates the complement system via the alternative (properdin) pathway, contributing to the host's inflammatory cascade. Due to its potent biological activity and ubiquitous presence in Gram-negative bacteria, endotoxin detection and removal are critical considerations in: - Pharmaceutical Manufacturing: Ensuring the safety of injectable drugs, vaccines, and medical devices. - Biotechnology Research: Preventing contamination in cell cultures, protein purification, and in vivo studies. - Clinical Diagnostics: Diagnosing bacterial infections and septic conditions.
