Adenoviruses are non-enveloped, double-stranded DNA viruses that are widely distributed in nature and are a common cause of acute upper respiratory tract infections, such as the common cold. Since their discovery in 1953, 47 human adenovirus serotypes have been identified, with three known to cause gastroenteritis. While most adenoviral infections are self-limiting and present with febrile illness involving inflammation of the respiratory tract and conjunctiva, certain serotypes have demonstrated oncogenic potential in experimental models. Adenoviruses are also known to establish latent infections, particularly in lymphoid tissues such as tonsils and adenoids, from which the virus can frequently be isolated following surgical removal. The duration of viral persistence and potential for long-term reactivation remain unclear, but reactivation has been documented in immunocompromised individuals, such as those with HIV/AIDS. The early gene product E1A is one of the first viral proteins expressed following infection and serves as a potent stimulator of cellular proliferation. E1A proteins, ranging from 35 to 46 kDa, are multifunctional and capable of overcoming the growth-inhibitory effects of TGF-β. They also form complexes with critical host proteins, including the retinoblastoma (Rb) tumor suppressor, thereby disrupting cell cycle control. While E1A expression alone can immortalize primary cells, full cellular transformation requires co-expression of the E1B region, which works synergistically with E1A to drive oncogenic progression.
